@EN The structure of the thyroglobulin may be affected by certain hormones, especially TSH. And there are some possibilities of the hormonogenic function of the thyroglobulin may be altered by the influence of certain hormones. So we have
investigated
the effects of different hormone conditions including No hormone condition, only TSH condition, and four hormone condition (TSH, insulin, hydrocortisone, transferrin) on the functional structure of the thyroglobulin secreted from the
monolayer-cultured
porcine thyroid cells.
Thyroglobulin was purified from the culture-medias of the monolayer-cultured porcine thyroid cells in different hormone conditions through the sephacryl s-300 column, and the purified thyroglobulin was iodinated by in-vitro addition of
carrier-free
NaI125 with lactoperoxidase and glucose oxidase. The iodinated thyroglobulin was digested with pronase and underwent paper chromatography in colloidin-NH4OH, and finally the bound I 125 to to iodoprotein (MIT, DIT, T3, T4) were estimated by gamma
counter.
The amount of the thyroglobulin secreted in medias was higher in order of 4H, TSH and NH. The iodine binding capacity of the thyroglobulin was higher in order of 4H, TSH and NH, especially in 4H with KI 2 nM/L significantly. There was no
significant
correlation between the iodoproteins in thyroglobulin and the hormone conditions. But the T3/T4 production rate in the thyroglobulin was greater in order of 4H, TSH and NH, especialy in 4H with KI 2 nM/L, significantly.
In conclusion, there are some possibility that the hormones including TSH, insulin, hydrocortisone, transferrin may alter the hormonogenic structure of the thyroglobulin, but further study is needed (J Kor Soc Endocrinol 8:308~315, 1993).
